The cAMP Hunter ADORA3 Gi cell line is a stable, engineered cell line for use in studying drug candidates (small molecules or biologics) targeting GPCRs. This cell line enables assessment of ligand (e.g. adenosine, NECA) based activation of ADORA3 GPCR activity via detection of decreasing levels of intracellular cAMP. This product uses EFC technology. The cells overexpress wild type ADORA3, a naturally Gi/Go coupled GPCR. Upon activation of ADORA3, the decrease in levels of intracellular cAMP leads to a decreased signal from EFC when the cAMP detection kit is applied. The included cell line overexpresses PTGER1, a naturally Gq coupled GPCR. ChemiBrite cells also express a clytin variant, allowing for luminescent as well as fluorescent signal detection. Human peripheral blood monocyte-derived immature dendritic cells are derived from negatively selected monocytes. Non-monocytes are labeled and depleted from the peripheral blood mononuclear cell population using immunomagnetic particles leaving purified, untouched monocytes. Untouched monocytes are cultured for 4-5 days in culture medium containing 10% FBS plus GM-CSF and IL-4. After culture, cells are checked for the expression of CD11c, HLA-DR, and CD83 and the lack of CD14 expression by flow cytometry.
